Genentech, a wholly owned member of the Roche Group, is constructing its first East Coast manufacturing facility at 4001 Friendship Road in Holly Springs, Wake County, North Carolina. The 700,000-square-foot biomanufacturing site will produce next-generation metabolic medicines, including obesity treatments, using advanced automation, robotics and digital manufacturing. Genentech first announced more than $700 million on May 12, 2025 and broke ground August 25, 2025. Its January 2026 announcement more than doubled the total site commitment to approximately $2 billion; AFT records that superseding total once and does not add the original amount. The August 2026 topping-out update confirms continued construction, with operations targeted for 2029. The project supports more than 500 high-wage manufacturing roles and more than 1,500 construction jobs; AFT records the conservative 500 permanent-manufacturing-job floor and excludes construction employment, incentives and Roche/Genentech's $50 billion national commitment.

North Carolina and Indiana remain major centers of investment, while new facilities are being planned across the South.
